[目的]探讨成纤维细胞体外成骨的诱导因素,体外获取大量成骨型成纤维细胞,为骨组织修复提供种子细胞。[方法]分离、纯化肉芽组织成纤维细胞,使其生长在含生肌液60g/L、FN40mg/L的条件培养液中。采用生物化学、组织化学和显微观察的方法,检测成纤维细胞的成骨标志。[结果]生肌液和FN促进成纤维细胞增殖、胶原合成、骨钙素分泌;生肌液可使成纤维细胞由梭形向圆形或多突形转化,细胞重叠成多层结构,其表面有钙盐堆积,逐渐呈云雾状物质,不断融合扩大形成不透光的结节,经趋骨性四环素特异标记证明为新生骨组织。[结论]生肌液可诱导成纤维细胞成骨。 |

Osteogenetic process of fibroblast in vitro induced by Shengjiye |

Abstract: |
[Objective] To explore the inducing factor of osteogenetic process of fibroblast in vitro. [Method] The fibroblasts were isolated and purified from grandlation tissue of New Zealand rabbit and were grown in culture madia containing Shengjiye 60g/L and Fibronectin(FN) 40mg/L.The osteogenetic features of fibrolast were investigated by biochemistry,histochemistry and microscopic observation. [Result] It was found that Shengjiye and FN could stimulate fibroblast to pro-liferate,secrete osteocaline and to synthetize collagen fibrils.The morphological observation found that the fibroblasts transferred from spindle shape to round or multiple,increased in number,confluenced and formed multilayered structure.They were piling up with calcium crystals on their surface and merging gradually into foggy substance.The foggy sub-stances were increasing and forming nodules,which were demonstrated to be new bone by histochemical study with specific labeling of tracycline fluorescence. [Conclusion] Shengjiye can induce the fibroblast in osteogenetic process. |
