不同治则中药单体对体外培养神经干细胞分化的影响

摘要:

[目的]研究不同治则中药对体外培养大鼠神经干细胞分化及成熟的影响。[方法]体外培养胎鼠神经干细胞,按适当浓度接种于24孔板,以β-tublin代表非成熟神经元marker,以tau代表成熟神经元marker,RT-PCR方法检测β-tublin、tau的表达,观察黄芩苷、栀子苷,黄芪甲苷、三七总皂苷对神经干细胞分化及成熟的影响。[结果]结果表明,黄芩苷、栀子苷能诱导神经干细胞特异性分化成神经元,而联合应用黄芪甲苷、三七总皂苷能进一步促进分化神经元的成熟。[结论]不同治则中药,针对神经再生的增殖、分化过程,联合用药,具有促进神经再生的作用。

Abstract:

[Objective] To investigate the influence of some extracts from Chinese herbs in different therapeutic principles on the differentiation and maturation of neural stem cell.[Methods] Neural stem cell was isolated from fetal rats rats.The cells were passaged three times.The cultured rat neural stem cells were plated on poly-D-lysin coated 24-well culture plates.β-tubin was used as the marker of immaturity neuron,tau as the marker of maturity neuron. The expression of β-tubin and tau mRNA were analyzed with RT-PCR method.The influence of baicalin,jasminoidin,Astragaloside IV Notoginsenoside extracted from Chinese herbs in different therapeutic principles on the differentiation and maturation was investigated.[Results] Baicilin and jasminoidin could induce neural stem cells differentiated to neuron specificity,while combined with Astragaloside IV and Notoginsenoside could promote the maturation of the differentiated neuron.[Conclusion] The comined use of Chinese herbs in differentiated therapeutic principles can promote the neuranagenesis.
