解毒祛瘀法对肿瘤血管生成影响的实验研究

摘要:

[目的]证实中医解毒祛瘀法能够抑制血管生成,调控血管生长基因,抑制肿瘤生长和转移作用。[方法]1)采用鸡胚绒毛尿囊膜(CAM)模型,将含有解毒祛瘀法提取液的载体置于7d胚龄的CAM上,作用48h观察抑制血管生成情况,并予氢化可的松相比较。2)采用家兔角膜移植肿瘤模型观察解毒祛瘀法对肿瘤血管生成抑制作用。3)解毒祛瘀法对荷瘤小鼠肿瘤生长及血管生成抑制作用研究。[结果]解毒祛瘀法能够使CAM及家兔角膜移植瘤的血管生成明显减少甚至消失。抑制荷瘤小鼠肿瘤生长,抑瘤率为49.6%,降低血管密度,促进肿瘤血管内皮细胞凋亡。[结论]中药复方解毒祛瘀法能够抑制CAM、家兔角膜移植瘤及荷瘤小鼠肿瘤生长和血管生成作用,提示解毒祛瘀法能够通过抑制肿瘤血管生成起到抑制肿瘤生长作用。

Abstract:

[Objective] To demonstrate whether Jiedu Quyu (relieve toxicity and remove blood stasis) can inhibit, regulate angiogenesis gene expression, inhibit the growth and metastasis of tumor.[Methods] 1)The chicken embryo chronic membrane (CAM)was used. The inhibitory effect was observed and compared with that of hydrocortisone 48 hours later. 2)Using rabbits with tumor induced by corneal transplanation the inhibitory effect of Jiedu Quyu on angiogenesis was observed in tumor-loaded mice. [Results] Jiedu Quyu could significantly decrease, even dispear, the angiogenesis in CAM or rabbits. The inhibitory rate in tumor-loaded mice was 49.6%.It could lower the vascular density and accelerate the apoptosis of vascular endothelial cells in tumor.[Conclusion] Complex Jiedu Quyu medicine in TCM can inhibit the growth and angiogenesis of tumor in CAM, rabbits with tumor of corneal transplantation and mice loaded with tumor, suggesting Jiedu Quyu method can inhibit the growth of tumor through the inhibition of angionenesis.
