[目的]研究真核生物翻译起始因子EIF3-P66在快速老化模型鼠(SAM-P/10)中的表达状况和针刺的调节作用。[方法]采用差别显示(DDRT-PCR)技术,分析SAM-P/10鼠伴加龄脑组织基因表达的变化及针刺的干扰作用。[结果]真核生物翻译起始因子EIF3-P66的表达伴增龄发生规律性的变化,伴快速老化表达增加,而针刺可下调其表达。同源性分析表明该序列与小鼠及人的翻译起始因子EIF3-P66高度同源。[结论]SAM-P/10的快速老化可能与EIF3-P66的异常表达有关,而针刺延缓衰老的可能机制之一是通过改善EIF3-P66的异常表达,继而减少蛋白质合成异常而发挥作用。 |

Abstract: |
[Objective] To explore the aging-associated genes of senescence accelerated mouse SAM and their relation to acupuncture. [Methods] The expression levels of cerebral genes of SAMP/10 with aging and acupuncture interference with aging were analyzed by applying DDRT-PCR technique. [Results] The expression level of a gene was observed to increase regularly with aging and revealed tendency to decrease after acupuncture points. Homology searching in the GenBank showed the fragment of the gene shares high homology with EIF3-P66 of mouse and human. [Conclusion] The accelerated aging of SAMP/10 is probably related to the abnormal over expression of EIF3-P66 and one of possible mechanism of acupuncture delaying the progress of aging is improve the abnormal synthesis of protein by down-regulating of its over expression. |
