[目的]研究解毒化瘀Ⅱ方防治急性肝衰竭肝性脑病的作用机制。[方法]将84只大鼠随机分为7组:空白组、阳性对照组、解毒化瘀Ⅱ方低、中、高剂量组、乳果糖组、安宫牛黄丸组。检测各组大鼠血清血氨(NH3)、支链氨基酸(BCAA)、芳香氨基酸(AAA)水平,脑线粒体钠钾ATP酶(Na+-K+-ATPase)、钙镁ATP酶(Ca2+-Mg2+-ATPase)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)、钙离子(Ca2+)含量。[结果]与模型组比较,解毒化瘀Ⅱ方能降低急性肝衰竭肝性脑病大鼠血清NH3的水平,恢复BCAA/AAA比值,提高脑线粒体Ca2+-Mg2+-ATPase、Na+-K+-ATPase、SOD的活力,抑制MDA生成,维持脑线粒体内Ca2+稳态,并以高剂量组最为显著。[结论]解毒化瘀Ⅱ方防治肝性脑病的机制可能与降低NH3、恢复BCAA/AAA比值、保护脑线粒体功能有关。 |

[Objective] To observe the mechanism of detoxification and removing Blood Stasis Formula II in preventing and treating hepatic encephalopathy in rats with acute hepatic failure(AHF).[Methods] Eighty-four rats with acute hepatic failure induced by injecting TAA were divided into seven groups:control group,treatment group,detoxification and removing blood stasis formula II groups of low,middle,high doses.lactulose group and angong niuhuan wan(Bezovr pill for miscarriage prevention)group.The level of serum NH3,BCAA,AAA,the activity of Na+-K+-ATPase,Ca2+-Mg2+-ATPase,SOD and the content of Ca2+ and MDA in cerebral mitochondrion were determined.[Results] The detoxification and removing blood stasis formula II was able to notably reduce the level of NH3,recover the ratio of BCAA/AAA,increase the activity of Ca2+-Mg2+-ATPase,Na+-K+-ATPase and SOD,inhibit the production of MDA and remain Ca2+ homeostasis in encephalic mitochondrion.The effect of high dose group is better than lactulose and the Chinese medicine angong niuhuang pill for miscarriage prevention groups(P<0.05).[Conclusions] The detoxification and removing blood stasis formula II can protect hepatic encephalopathy in rats with acute hepatic failure.Its mechanism might be due to reducing the content of NH3,recovering the ratio of BCAA/AAA,protecting the function of cerebral mitochondrion. |
