[目的]建立银屑灵涂膜剂中5种蒽醌类成分的含量测定方法。[方法]采用高效液相色谱法(HPLC),使用Irregular-HC18(250mm×4.6mm10μm)色谱柱,流动相为甲醇-0.1%磷酸水溶液(85:15),流速:1.00mL/min,检测波长:254nm。[结果]芦荟大黄素的线性范围为0.784~15.68μg(r=0.9994,n=6),平均回收率为100.37%,峰面积(RSD)为1.31%;大黄酸的线性范围为0.748~14.96μg(r=0.9996,n=6),平均回收率为100.12%,RSD为1.60%;大黄素的线性范围为0.804~16.08μg(r=0.9998,n=6),平均回收率为101.20%,RSD为1.79%;大黄酚的线性范围为0.806~16.12μg(r=0.9999,n=6),平均回收率为100.64%,RSD为1.20%;大黄素甲醚的线性范围为0.466~9.32μg(r=0.9999,n=6),平均回收率为101.89%,RSD为0.54%。[结论]HPLC法准确、可靠、简单,可用于银屑灵涂膜剂中蒽醌类成分的含量测定。 |

Abstract: |
[Objective] To establish a method for determining the contents of five kinds of anthraquinones components in Yinxieling(treating psoriasis) Film. [Methods] HPLC was carried out using a irregular-H C18 column (250 mm×4.6 mm 10 μm) and taking methanol-0.1% phosphoric acid(85:15) as the mobile phase with flowing rate at 1.00 mL/min and detecting at 254 nm.[Results] The calibration curve of aloe emodin was linear from 0.784 μg to 15.68 μg (r=0.999 4,n=6) and the average recovery was 100.37 % with RSD 1.31%. The calibration curve of rhein was linear from 0.748 μg to 14.96 μg(r=0.999 6,n=6)and the average recovery was 100.12 % with RSD 1.60 %. The calibration curve of emodin was linear from 0.804 μg to 16.08 μg (r=0.999 8,n=6)and the average recovery was 101.20 % with RSD 1.79 %. The calibration curve of chrysophanol was linear from 0.806 μg to 16.12 μg (r=0.999 4,n=6)and the average recovery was 100.64% with RSD 1.20%. The calibration curve of physcion was linear from 0.46 μg to 9.32 μg(r=0.999 9,n=6)and the average recovery was 101.89% with RSD 0.54%.[Conclusions] This method is proved to be accurate,reliable and simple and can be used for the determination of anthraquinones components in Yinxieling Film. |
