摘要: |
[目的] 通过复制动静力失衡性颈椎病模型大鼠,经免疫组化及分子生物学法检测颈椎间盘相关炎症因子,以探讨"四天穴"针法对颈椎病外周炎症病理网络链中关键环节(包括炎性因子、疼痛、缺血缺氧、自由基释放等)的影响及其作用机制。[方法] 采用上海中医药大学脊柱病研究所的经典造模方法,重复建立动静力失衡性颈椎病模型,经3个月诱导,该模型大鼠出现颈椎间盘退变,制成颈椎病模型。60只大鼠共分为4组(假手术组、模型组、夹脊穴针刺组、四天穴针刺组),15只/组,经针刺干预后,于1、2、4周3个时间点,每次每组处死5只大鼠,后用免疫组化染色法(SABC)观察对椎间盘中白细胞介素-1β(IL-1β)、中环氧合酶(COX-2)、诱导型一氧化氮合酶(iNOS)蛋白表达的影响,分子生物学逆转录-聚合酶链式反应(RT-PCR)法观察对椎间盘中IL-1βmRNA及iNOS mRNA表达的影响。[结果] 四天穴针刺组与夹脊穴针刺组均可降低模型大鼠颈椎间盘内IL-1β、COX-2、iNOS的蛋白表达,降低IL-1βmRNA、iNOS mRNA的表达。与模型组比较,四天穴针刺组及夹脊穴针刺组在1、2、4周3个时间相中,均有显著性差异(P<0.05);四天穴针刺组间比较,在1、2、4周3个时间相中,4周组均极显著低于1周组(P<0.01);两针刺组间比较,部分存在明显差异。提示"四天穴"针法对颈椎病外周炎症病理网络链中炎性因子表达、根性痛、缺血缺氧、自由基损伤等关键环节均有干预作用。[结论] "四天穴"针法能够减缓颈椎间盘退变的程度,其作用机制与减少或抑制椎间盘退变后外周炎症病理网络链中关键环节的表达密切相关。 |

Effect of acupuncture of Sitian Points on peripheral inflammatory factors in pathologic net in rats with experimental move-static power unbalanced disease of vertebrae cervicales |

Abstract: |
[Objective] To study the effect and mechanism of Acupuncture on Sitian Points (ASTP) on the peripheral inflammatory factor in vertebrae cervical syndrome (CS). We induced the imbalance of dynamic and static forces in the cervical disk in Wistar rats, examined the expression of IL-1β, COX-2, iNOS by immunohistochemical method and the expression of IL-1βmRNA, iNOSmRNA by molecular biology method.[Methods] The model of cervical disc dynamic and static forces imbalance was established with the methods used in Shanghai University of TCM, which was similar to the pathogenesis of CS. Sixty Wistar rats were randomly divided into 4 groups with 15 each: SO group, model group, APJJX group, APSTX group. After acupuncture, every group was randomly divided into 3 time groups again with 5 each: the groups of 1, 2, 4 weeks. Then we investigated the effect of APSTX from the followed aspects: to observe the influence on the expression of IL-1β, COX-2, iNOS in intervertebral disc by immunohistochemical SABC method; to observe the influence on the expression of IL-1βmRNA, iNOSmRNA in intervertebral disc by molecular biology RT-PCR method.[Results] APSTP can reduce the expression of COX-2, IL-1β, iNOS and the expression of IL-1βmRNA, iNOSmRNA in the degenerated intervertebral disc of CS model rats through inhibiting the effect of peripheral inflammatory factor.[Conclusions] Puncturing APSTP can markedly delay the process of degenerated intervertebral disc in CS rats. The mechanism of action may be correlated with reducing the effect of peripheral inflammatory factor. |
