益智宁神颗粒对自发性高血压模型大鼠脑组织去甲肾上腺素、多巴胺的影响

摘要:
[目的]探讨益智宁神颗粒治疗儿童注意缺陷多动障碍(ADHD)的神经生化机制。[方法]将48只自发性高血压(SHR)大鼠随机分为6组,益智宁神颗粒高、中、低剂量组,静灵口服液组,利他林组,模型组。将8只同龄Wistar大鼠设为正常对照组。通过高效液相-电化学法,检测各组大鼠脑组织中多巴胺(DA)、去甲肾上腺素(NE)、高香草(HVA)及其代谢产物含量。[结果]模型组大鼠脑组织中DA、NE含量显著高于正常对照组,HVA差别不明显。高剂量益智宁神颗粒可降低模型大鼠脑组织中DA、NE的含量,并使两者处于平衡状态,其疗效优于利他林及静灵口服液。中、低剂量益智宁神颗粒、利他林及静灵口服液对大鼠脑组织中DA、NE含量无显著影响。[结论]益智宁神颗粒抑制模型大鼠多动、探索行为,改善认知功能的作用,可能与降低脑组织中DA、NE含量,并使其处于平衡状态有关。

Effect of Yizhiningshen granule on NE, DA, HVA in brain tissue of SHR rats

Abstract:
[Objective] To explore the nervobiochemical mechanism of Yizhiningshen granule in treating Children attention deficit hyperactivity disorder(ADHD). [Methods] The treatment group was given the high, middle and low dosage of Yizhiningshen granule; oral Jingling liquid was given in control group of TCM and Ritalin in control group of Western medicine. To determine the DA, NE, HVA level and its metabolic product HPLC and electrochemistry method were used. [Results] The DA and NE content of model group were obviously higher than that of normal control group and the HVA content of model group wasn’t obviously different in brain tissue. Yizhiningshen granule could effectively decrease the DA and NE content of model rats and could regulate the balance of the DA and NE content. The treatment effect of Yizhiningshen granule had an obvious advantage when compared with that of Ritalin and Jingling liquid for oral use. [Conclusions] Yizhiningshen granule may inhibit restlessness and exploratory behavior of model rats and improve recognition function. The effect was concerned with decreasing the DA and NE content of brain tissue and regulating the balance state of the DA and NE content.
