反相高效液相色谱法同时测定不同产地夏天无药材中原阿片碱和延胡索乙素的含量 |

摘要: |
[目的]建立了反相高效液相色谱(RP-HPLC)法同时测定夏天无药材中原阿片碱与延胡索乙素含量的方法,并对不同产地夏天无药材中原阿片碱与延胡索乙素含量进行了考察。[方法]采用kromasilC18色谱柱(250mm×4.6mm,5μm);以乙腈-0.1%磷酸溶液(pH=6.5)为流动相连续梯度洗脱,流速1.0mL/min;检测波长280nm,柱温35℃。[结果]原阿片碱的回归方程为Y=823332.1X+9694.4,r=0.9997(n=5),线性范围为0.22~1.10μg;延胡索乙素回归方程为Y=1047499X-15305.6,r=0.9996(n=5),线性范围为0.174~0.870μg。样品的平均回收率分别为98.9%,99.7%,峰面积(RSD)分别为1.6%,1.8%(n=5)。[结论]此法简便、准确、快速,可用于药材中延胡索乙素与原阿片碱含量的测定,也可用于含这两种生物碱的药材的质量评价;两种生物碱含量在不同产地及来源夏天无药材中有较大差别。 |

[Objective] A RP-HPLC method is established for simultaneous determination of the content of protopine and tetrahydropalmatine in Corydalis decumbens (Thunb.) Pers from the different habitats. [Methods] The chromatographic separation was performed on a Kromasil C18 column with a linear gradient elution of acetonitrile-0.1% phosphoric acid was used. The detection wavelength was set at 280 nm; flow rate was 1.0 mL/min and column temperature was set at 35 ℃. [Result] Protopine and tetrahydropalmatine had a good linearity in the range of 0.22~1.10 μg and 0.174~0.870 μg,The average recoveries of this method were 98.9% and 99.71%;RSD were 1.6% and 1.8%(n=5)respectively and detected at wavelength of 280 nm. [Conclusion] The contents of the two alkaloids in Corydalis decumbens (Thunb.) Pers. from the different habitats show obvious difference. The method is simple,accurate,quick and suitable to determine content and evaluate quality for the herds. |
