[目的]探讨参附注射液和生脉注射液对心肌细胞Ca2+-ATP酶的影响机制,旨在为参附注射液和生脉注射液的临床应用提供实验依据。[方法]采用胰酶消化法体外培养大鼠乳鼠心肌细胞,通过差速贴壁的方法纯化细胞后,分别把参附注射液和生脉注射液选取3个浓度梯度直接作用于对数生长期的心肌细胞,药物作用2h后检测其对心肌细胞内Ca2+-ATP酶活力的影响。[结果]参附注射液的低、中、高剂量均能使心肌细胞内Ca2+-ATP酶活力增强,以中剂量最为明显(P<0.05);生脉注射液低、中剂量对心肌细胞内Ca2+-ATP酶活力的影响较小,高剂量有增强心肌细胞内Ca2+-ATP酶活力的趋势,但没有统计学意义。[结论]提示参附注射液对心脏的作用机制可能与其能提高心肌细胞内Ca2+-ATP酶活力有关;生脉注射液对心肌细胞内Ca2+-ATP酶活力有一定影响,但其可能主要是通过其他机制作用于心肌细胞。 |

Abstract: |
[Objective] To explore the effect of Shenfu injection and Shengmai injection on Ca2+-ATPase of myocardial cell in mice and provide the basis for the clinical application. [Methods] Using trypsin digestion the myocardial cells were cultured in vitro. After purifying the cells with adhering to the wall with different velocity Shenfu injection and Shengmai injection with three concentrations gradient were added directly to the myocardial cells in logarithmic developing period. Two hours later its effects on the activity of Ca2+-ATPase in myocardial cells was detected. [Results] Shenfu injection of the low, middle and high concentration could increase the Ca2+-ATPase activity of myocardial cells, and the effect of middle concentration was the most obvious. (P<0.05) Lower effect of low and middle concen-tration of Shengmai Injection was found. The high concentrations of Shenfu Injection, however, developed a increasing trend of the Ca2+-ATPase activity. [Conclusion] It is indicated that the effect of Shenfu injection on the heart may be related to the increasing of Ca2+-AT-Pase activity in myocardial cells;Shengmai injection has some effect on Ca2+-ATP activity in myocardial cells to a certain extent through other mechanisms on the myocardial cells. |
