P450arom、CYP19及TGF-β1在雄性肾阳虚不育大鼠睾丸中的表达

摘要:
[目的]观察限速酶芳香化酶(P450arom)、芳香化酶基因(CYP19)和转化生长因子(TGF-β1)在腺嘌呤诱导的雄性肾阳虚不育大鼠睾丸中的表达,探讨肾虚引起的雄性不育的病理机制。[方法]20只雄性大鼠随机分为4组:正常对照组,模型Ⅰ、Ⅱ、Ⅲ组,5只/组。用腺嘌呤100、150、200mg(/kg·d),分别给模型Ⅰ,Ⅱ,Ⅲ组大鼠灌胃30d,诱导肾阳虚不育模型。用免疫组化浓缩型免疫组化染色法(SABC法),测定P450arom、CYP19和TGF-β1在实验大鼠睾丸中的表达。[结果]CYP19在大鼠睾丸的间质细胞、长形精子细胞中均有表达;P450arom和TGF-β1在睾丸的间质细胞有表达;其免疫阳性物质均位于细胞质内。模型组大鼠睾丸中TGF-β1的表达明显高于正常对照组(P<0。05),而CYP19、P450arom的表达明显低于正常对照组(P<0。05);各模型组之间,TGF-β1、CYP19、P450arom的表达无显著性差异(P>0。05)。[结论]P450arom、CYP19及TGF-β1在睾丸表达的异常可能与肾虚引起的雄性大鼠不育有关。

Abstract:
[Objection] To study the expression of P450arom,CYP19 and TGF-β1 in the testis of infertile rats with adenine-modeled kidney-yang deficiency and the pathologicalmechanian of infertility with kidney-yang deficiency.[Methods] Twenty-three month male SD rats were divided randomly into four groups with five in each group(normal control,model,Ⅰ,Ⅱ and Ⅲ).The experimental rats had been fed with adenine [100,150,200(mg/kg·d)] in 30 days and the expression levels of P450arom,CYP19,TGF-β1 were measured with immunohistochemical SABC method.[Results] CYP19 was expressed in cell of Leydig,dolicho-nematoblast in rats’testis;P450arom and TGF-β1 were positive expressed in cytoplasm of Leydig’s cell;with their positive immuno-responsive substance locating in the cytoplasm.Compared with the normal control,TGF-β1 expression was higher in kidney-yang deficiency group(P<0.05);CYP19 and P450arom expression were higher in the kidney-yang deficiney group(P<0.05).Among the three model groups,the expression of TGF-β1,P450arom,CYP19 had no significant deviation(P>0.05).[Conclusion] The abnormality expression P450arom,CYP19,TGF-β1 in testis was possible with infertility with kidney-yang deficiency,for further approach P450 arom,CYP19,TGF-β1in infertility rats with kidney-yang deficiency gonepoiesis and correlation provided preliminary experimental evidence.
