[目的] 研究针刺对急性心肌缺血大鼠缺血心肌基因表达谱的影响.[方法] 建立左前降支冠状动脉结扎大鼠心肌缺血模型,分组干预后分离提取缺血心肌总RNA,与大鼠全基因表达谱芯片杂交后进行检测,对筛选得到的针刺治疗过程中的差异表达基因,利用基因功能分类体系寻找和分析显着富集差异表达基因的复合功能分类,从基因功能水平探索针刺对急性心肌缺血模型大鼠缺血心肌基因表达谱的影响.[结果] 发现心肌缺血24 h后表达上调的398条基因中,经过间断针刺内关穴3次,24 h后其中298条基因(占表达上调基因的74.87%)的表达出现不同程度的下调趋势;心肌缺血24 h后表达下调的338条基因中,经过间断针刺内关穴3次,24 h后其中188条基因(占总下调基因的55.62%)的表达出现不同程度的上调趋势,表明针刺内关穴对缺血心肌差异表达基因有良性调节趋势.针对筛选出来的差异表达基因蛋白磷酸酶1B(Ppm1b)、琥珀酸辅酶A连接酶(Suclg1)、谷氨酸草酰乙酸转氨酶2(Got2)、苹果酸脱氢酶(Mdhl)、丙酮酸脱氢酶磷酸酶同工酶(PDP1)、延胡索酸水合酶(Fh)、羟肽酶b2(Cpb2)、酪氨酸单氧化酶激活蛋白酶(Ywhaz)、S100钙结合蛋白a9(S100A9)、Na+/K+转运ATP酶B1亚基(Atp1b1)、肌钙蛋白I(TnI)、肌球蛋白轻链激酶3(Myl3)、肌集钙蛋白2(Casq2)、肌红蛋白(Mb),其功能模块主要涉及氧化磷酸化、线粒体电子传递、电子传递偶联三磷酸腺苷(ATP)合成、能量代谢、磷酸盐代谢过程、单价无机阳离子转运、辅酶代谢过程等.[结论] 针刺内关穴可能是通过影响上述基因,从而对急性心肌缺血发挥保护作用. |

Effect of acupuncture at Neiguan acupoint on the gene expression profile of rats with acute myocardial ischemia |

Abstract: |
[Objective] To study the effect of acupuncture on the gene expression profile in acute myocardial ischemia. [Methods] Aacute myocardial ischemic was induced by ligating the left anterior descending branch of coronary artery, and the total RNA of the ischemic myocardium was extracted after grouping and intervention. Afterwards, we hybridized the extraction with the whole gene expression profile chip in rat and detected them. In the differential expressed gene during therapeutic process after screening, we searched and analyzed the compound functional classification of the significant aggregative differential expression by the means of gene functional classification system and discussed the effect of acupuncture on the gene expression profile in acute myocardial ischemia from the level of gene function. [Results] The expression of 398 genes was up-regulated after 24 hours of myocardial ischemia. However, the expression of 298 gene (74.87% of the total up-regulated genes) showed the tendency of down-regulation after acupuncture at Neiguan acupoint three times intermittently; in the 338 down-regulated genes after 24 hours of myocardial ischemia, the expression of 188 genes (55.62% of the total down-regulated genes) showed the tendency of up-regulation after acupuncture at Neiguan acupoint three times intermittently, indicating a benign regulative action of the expression of ischemia myocardium gene after acupuncture at Neiguan acupoint. In the screened differential expression genes, the functional module of Ppm1b, Suclg1, Got2, Mdhl, PDP1, Fh, Cpb2, Ywhaz, S100A9, Atp1b1, TnI, Myl3, Casq2, Mb, mainly involved the oxidative phosphorylation, the electron transfer of mitochodria, the electron transfer coupling of ATP synthesis, the energy metabolism, the process of phoshates metabolism, the univalent inorganic cation transfer, the process of coenzyme metabolism and so on. [Conclusion] Acupuncture at Neiguan acupoint may protect the myocardium by influencing the genes mentioned above. |
