转运蛋白是一种允许有机和无机溶质高效率和选择性跨膜透过的膜蛋白。转运蛋白基因的分子识别即通过克隆cDNA外源表达系统的体外实验方法, 阐明转运蛋白的功能。代谢组学分析是通过定量比较某化合物在野生型和转运蛋白基因敲除型小鼠的尿液、血液或组织样本的信息, 从而揭示生物体内转运蛋白的真正底物。这种方法也可以用来发现孤儿转运蛋白功能。举例说明基于毛细管电泳与质谱联用(CE-MS)技术, 应用代谢组学分析, 揭示了SLC22转运蛋白家族中一个孤儿转运蛋白的功能。 |

Application of metabolomics in the analysis of in vivo function of transporters |

Abstract: |
Transporters are membrane proteins that allow efficient and selective permeation of organic and inorganic solutes through the membrane. The molecular identification of transporter genes has facilitated the understanding of functions of transporter proteins by in vitro experimental approaches in which cloned cDNAs are expressed in exogenous expression systems. The metabolomics analysis to compare chemical compounds in urine, blood or tissue samples quantitatively between wild type and transporter-knockout mice gives us clues to the real substrates of the transporter in vivo. This approach can also be used to find out functions of orphan transporters. For example, we have applied metabolomics with capillary electrophoresis-mass spectrometry (CE-MS) to reveal the function of an orphan transporter in SLC22 transporter family. |
