火针干预脊髓损伤大鼠后血清对神经干细胞增殖分化的影响

摘要:
[目的] 研究离体状态下火针干预脊髓损伤大鼠后提取血清对神经干细胞增殖分化的影响。[方法] 将15只SD大鼠随机分为假手术组、模型组及火针组,相应干预3 d后取各组大鼠血清。从孕14天昆明小鼠胚胎脊髓组织中分离培养神经干细胞,同时采用上述各组血清进行培养,7 d后观察各组神经干细胞增殖与分化情况。[结果] 神经干细胞在各组血清中均能存活、增殖并可向神经元分化,火针组神经干细胞增殖数目显着多于假手术组及模型组,且火针组神经元分化比例高于其余两组(P<0.05).[结论] 火针干预脊髓损伤大鼠后血清可促进神经干细胞增殖并向神经元分化。

Abstract:
[Objective] To study the influence of the extracted serum of SCI rats after fire needle intervention on the proliferation and differentiation of neural stem cells in vitro. [Methods] Fifteen SD rats were randomly divided into sham operation, model and fire-needle groups, and the serum was extracted in each group after 3 days' corresponding intervention. The neural stem cells were separated from the spinal cord tissue in Kunming mice embryo after 14 days' pregnancy, and then cultured by all the above serum in each group to observe the proliferation and differentiation states of neural stem cells 7 days later. [Results] Neural stem cells of each group could survive, proliferate and differentiate into neurons. The proliferation number of neural stem cells in fire needle group was obviously more than that in sham operation and model group, and the percentage of differentiation into neurons in fire group was higher than that in the other groups (P<0.05). [Conclusions] The serum of SCI rats after fire needle intervention could promote the proliferation and differentiation into neurons of neural stem cells.
