[目的] 选择适合的评价方法及纳入排除标准,以提高对脑缺血模型是否成功的准确判定程度。[方法] 采用线栓法建立Wistar大鼠的大脑中动脉阻塞(MCAO)模型,用激光多普勒分别测量造模前、后局部脑血流量(rCBF).分别在缺血后1、2、3、6、9、12、15、18、21、24 h进行Longa神经行为学评分,断头取脑,利用2,3,5-氯化三苯基四氮唑(TTC)染色法对脑梗死情况进行判定。[结果] 经解剖和TTC染色验证,神经行为学评分法对44只MCAO大鼠模型判读准确,准确率为75.86%,结合解剖结果,准确率为86.21%.多普勒脑血流量检测法对51只模型MCAO大鼠模型判读准确,准确率为87.93%.结合解剖结果,准确率为100%.[结论] 激光多普勒血流测量法对大鼠大脑中动脉梗死模型是否成功的判断具有良好的敏感性和准确性,尤其适用于超急性期大脑中动脉梗死模型的判定。 |

Discussion on evaluating criteria of model of middle cerebral artery occlusion in rats |

Abstract: |
[Objective] To choose criteria of suitable evaluation and enrolling and excepting method for improving the judgment accuracy of the cerebral ischemia model. [Methods] Adopting suture-occluded method the model of middle cerebral artery occlusion (MCAO) was established in Wistar ratsl. The change of rCBF was measured with Laser Doppler Blood Flow assessment. The neurological state was assessed by Zea-Longa neurological deficit scores at 1, 2, 3, 6, 9, 12, 15, 18, 21 and 24 h after cerebral infarction; then the samples were prepared and Triphenyltetrazolium chloride (TTC) staining was used to evaluate the infarct condition. [Results] Anatomic examination and TTC staining showed that neurobehavioral scoring method can correctly judge for 44 rats with MCAO and the accuracy rate was 75.86%. When combined with the result of the anatomic examination, the accuracy rate was rised to 86.21%. The measure of rCBF with Laser Doppler blood flow measurement was accurate in 51 rats, and its accuracy rate was 87.93%. When combined with the result of anatomic examination, the accuracy rate will be 100%. [Conclusion] Measuring the changing of rCBF with Laser Doppler blood flow assessment is a sensitive and accurate method to evaluate the MCAO in rats, especially used in the super-acute phase. |
