温补肾阳方对心肌梗死后心力衰竭大鼠血流 动力学及血清肌钙蛋白I和脑钠肽含量的影响 |

摘要: |
[目的] 研究温补肾阳方对心肌梗死后心力衰竭大鼠心脏血流动力学及血清心肌肌钙蛋白I(TNI)和脑钠肽(BNP)含量的影响。[方法] 采用结扎冠状动脉前降支术制备心肌梗死后心力衰竭大鼠模型, 造模成功后, 将大鼠随机分为模型组, 曲美他嗪为对照组, 假手术组为空白对照组, 温补肾阳方高、中、低剂量组。给药8周后测血流动力学指标, 如左室收缩压(LVSP)、左室舒张压(LVEDP)、左室内压上升下降最大速率(±LVdp/dtmax);应用双抗体夹心法测定心肌梗死后心力衰竭大鼠药物干预前后TNI水平;采用酶联免疫吸附法检测治疗前后大鼠BNP水平。[结果] 治疗后测血流动力学, 与模型组相比, 温补肾阳方中、高剂量组与曲美他嗪组均可显着升高心力衰竭大鼠LVSP和+LVdp/dtmax(P<0.01),降低其LVEDP、-LVdp/dtmax(P<0.01),且高剂量组要优于中剂量。与模型组相比, 补肾阳方高、中、低剂量组及曲美他嗪组均可显着降低心力衰竭大鼠TNI及BNP水平(P<0.01).[结论] 温补肾阳方能提高心力衰竭大鼠的心脏收缩、舒张功能, 减轻心肌受损, 改善心功能。 |

Abstract: |
[Objective] To observe the effect of Wenbu Shengyang (warming kidney Yang) prescription on cardiac hemodynamics, the content of cardiac troponin I(TNI) and brain natriuretic peptide(BNP) in rats with heart failure after myocardial infarction. [Methods] The heart failure was established in rats by ligating left anterial descending coronary artery. Rats were divided into model group, trimetazidinetriazine control group, sham operation normal control group, high, middle and low doses of warming kidney Yang prescription group randomly. After treatment for 8 weeks, we measured the hemodynamics including left ventricular systolic pressure (LVSP), left ventricular diastolic pressure(LVDP), the increased or decreased maximum rate of left ventricular pressure(±LVdp/dtmax). The level of TNI was determined through sandwich ELISA before and after medicine intervention in myocardical tissue. The content of BNP was determined by ELISA before and after treatment. [Results] Compared with model group, warming kindey Yang prescription with middle and high dosage and trimetazidinetriazine control group could obviously increase LVSP, +LVdp/dt(P<0.01) and decrease LVEDP, -LV dp/dt (P<0.01). The effect of high doses was better than middle doses. Compared with the model group, every doses of warming kidney Yang and trimetazidine could significantly lower the levels of TNI and BNP(P<0.01). [Conclusion] Warming kidney Yang prescription can significantly improve cardiac systolic and diastolic function of rats with heart failure as well as reduce myocardial damage and improve heart function. |
