| [目的] 建立液质联用方法同时检测给药后大鼠胆汁中木通皂苷D及其代谢产物,研究木通皂苷D在大鼠胆汁中的排泄情况。[方法] 大鼠灌胃给药木通皂苷D后按照设定时间点收集胆汁,通过建立的液相质谱联用方法分析胆汁中的木通皂苷D及其代谢产物的含量变化情况。[结果] 实验同时检测到了木通皂苷D与其3个代谢产物,并对浓度较大的木通皂苷D和HN saponin F进行了定量。木通皂苷D及HN saponin F在3.2~2 000 ng/mL与0.8~500 ng/mL范围内线性关系良好,日间日内精密度均小于7.72%,给药24 h内累积排泄量为5 845.5 ng与3 996.8 ng.[结论] 该方法符合生物样品测定要求,可用于胆汁中木通皂苷D及其代谢产物HN saponin F的定量分析。 |

| [Objective] To establish a method for simultaneous determination of asperosaponin VI and it's metabolites in rat bile by liquid chromatography-mass spectrometry (LC-MS/MS) and to study the excretion of asperosaponin VI from rat bile. [Methods] Bile samples were collected according to the designed time periods after intragastric administration. The content of asperosaponin VI and it's metabolites in rat bile were then analyzed using the established LC-MS/MS method. [Results] Asperosaponin VI and its three metabolites were simultaneously determined; asperosaponin VI and HN saponin were quantified at a higher concentration. Calibration curves were generated over a concentration range of 3.2~2 000 ng/mL for Asperosaponin VI and 0.8~500 ng/mL for HN saponin, respectively. Inter- and intra- day RSDs were both less than 7.72%. Biliary accumulated excretion for 24 hours after administration were 5 845.5 ng and 3 996.8 ng, respectively. This method is suitable for the determination of asperosaponin VI and its metabolite HN saponin F. [Conclusion] This method meets the requirements for the quantitative determination of biological samples and can be used for quantitative analysis of asperosaponin VI and its metabolite HN saponin F in rat bile. |
