HPLC法测定金黄膏中小檗碱和巴马丁的含量

摘要:
[目的] 建立同时测定金黄膏中盐酸小檗碱、巴马丁含量的方法。[方法] 采用高效液相色谱法,色谱柱为Apollo C18(250 mm×4.6 mm,5 μm),流动相为乙腈-0.1%磷酸(二元梯度洗脱),检测波长为 346 nm,流速为1.0 mL/min,柱温为30 ℃。[结果] 盐酸小檗碱和巴马丁的进样量分别在0.007 25~0.145 μg,0.005 5~0.110 μg范围内与各自峰面积积分值呈良好的线性关系,平均加样回收率分别为98.8%和100.9%.[结论] 该方法操作简便、重现性好,适用于金黄膏中巴马丁和盐酸小檗碱的含量测定。

Determination of berberine and palmatine in Jinhuang cream by HPLC

Abstract:
[Objective] To establish a method for simultaneous determination of berberine and palmatine in Jinhuang cream. [Methods] A HPLC method was performed on Apollo C18 (250 mm×4.6 mm, 5 μ) column. The mobile phase consisted of acetonitrile-0.1% phosphoric acid(gradient elution)with the detection wavelengh at 346 nm. The flow rate was 1.0 mL/min and the column temperature was at 30 ℃. [Results] The content of berberine hydrochloride in the range of 0.007 25~0.145 μg and the content of palmatine in the range of 0.005 5~0.110 μg had good linear relationships with the area of eluted peak,and the recovery rate(the average recoveries) was 98.8% and 100.9% respectively. [Conclusion] This method is simple and repeatable, and is suitable for the content determination of berberine and palmatine in Jinhuang cream.
