[目的] 探讨天麻钩藤饮对自发性高血压大鼠(SHR)的降压机制及对心肌组织B细胞淋巴瘤-2基因相关X蛋白(Bax)、半胱氨酸天冬氨酸特异性蛋白酶-3(Caspase-3)蛋白表达的影响。[方法] 20只14周龄自发性高血压大鼠(SHR)随机分为空白组6只、中药组7只、西药组7只。中药组灌服天麻钩藤饮20 mL/(kg·d),相当生药20g/(kg·d).西药组灌服雷米普利1 mg/(kg·d).空白组灌服同体积的纯净水。连续灌胃4周。分别于给药0、1、2、3、4周测量血压和心率。给药4周后,腹主动脉取血处死,用酶联免疫吸附实验法测定血清血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)含量,用Western blot方法检测心肌组织Bax及Caspase-3蛋白表达。[结果] 1)从给药3周后开始,与空白组比较,天麻钩藤饮能明显降低收缩压(P<0.05,P<0.01).天麻钩藤饮降压效果虽没有西药明显,但整体上能控制收缩压的升高,并有逐渐降低的趋势。2)天麻钩藤饮和西药雷米普利对心率的变化均没有明显影响。3)与空白组比较,天麻钩藤饮和西药均能明显降低血清的AngⅡ和ALD含量(P<0.01).4)与空白组比较,天麻钩藤饮和西药均能明显降低心肌组织的Bax、Caspase-3蛋白表达(P<0.01).[结论] 天麻钩藤饮能有效控制SHR大鼠收缩压的升高。天麻钩藤饮的降压机制可能与降低AngⅡ和ALD含量有关,并且还可能通过减少Bax、Caspase-3蛋白表达来抑制心肌细胞的凋亡,实现对心肌细胞的保护功能。 |

Effect of the Tianma Gouteng Yin regulating the protein expression of Bax and Caspase-3 of myocardial tissue in spontaneously hypertensive rats |

Abstract: |
[Objective] To investigate the mechanism of Tianma Gouteng Yin (TMGTY) reducing blood pressure and the protein expression of Bax and Caspase-3 in myocardial tissue in spontaneously hypertensive rats. [Methods] Twenty 14-week-old SHR rats were randomly assigned to control group (n=6), TCM group (n=7) and Western medicine group (n=7). The rats in TCM group were given TMGTY with the dose of 20 g/(kg·d); those in the western medicine group were given ramipril [1 mg/(kg·d)]; those in control group were given the same volume of pure water. Blood pressure and heart rate were measured weekly. After 4 weeks, abdominal aortic blood and sacrifice animals. Enzyme-linked immunosorbent assay was used to analyze the content of AngⅡ and ALD in serum. Western blot was used to analyze quantitatively Bax and Caspase-3. [Results] 1) Starting from the third week after the administration, compared with the control group, TMGTY significantly reduced systolic blood pressure (P<0.05, P<0.01). Although not significant antihypertensive effect, TMGTY controlled the overall increase in systolic blood pressure, and systolic blood pressure gradually decreased. 2) TMGTY and ramipril had no significant effect on heart rate. 3) Compared with the control group, TMGTY and ramipril significantly reduced the content of AngⅡ and ALD in serum (P<0.01). 4) Compared with the control group, TMGTY and ramipril significantly reduced the protein expression of Bax and Caspase-3 in myocardial tissue (P<0.01). [Conclusion] TMGTY could effectively control the increase in systolic blood pressure in SHR. The antihypertensive mechanism of TMGTY may be reducing ALD and AngⅡ. Further, by reducing the protein expression of Bax and Caspase-3, TMGTY possibly inhibited the apoptosis of cardiomyocytes, and achieved protection to cardiomyocytes. |
