[目的]研究藏红花素(Crocin)对实验性2型糖尿病大鼠睾丸组织并发症损伤的保护作用。[方法]100只SD大鼠按体质量随机分为正常对照组、模型对照组、藏红花素25 mg/(kg·d)、50 mg/(kg·d)、100 mg/(kg·d)组,每组20只,灌胃给药,疗程12周;采用链脲佐菌素(STZ)连续3 d腹腔注射30 mg/(kg·d)的方法制备实验性2型糖尿病大鼠模型。测定各组大鼠空腹血糖(FBG)、空腹胰岛素(FINS)和糖化血红蛋白(GHb)水平,称量大鼠体质量、睾丸质量后计算睾丸指数,测定血清睾酮含量;采用苏木精-伊红(HE)染色对睾丸组织进行病理学检查,通过透射电镜观察睾丸组织超微结构改变;末端脱氧核苷酰基转移酶介导dUTP切口末端标记法(TUNEL)观察睾丸组织细胞凋亡状况。[结果]经藏红花素50~100 mg/(kg·d)治疗12周,2型糖尿病大鼠FBG显著降低、FINS显著升高、GHb显著降低(P<0.05或P<0.01);体质量、睾丸指数和血清睾酮含量显著升高(P<0.05或P<0.01)。经藏红花素治疗12周,糖尿病大鼠睾丸组织曲细精管萎缩、生精细胞脱落、生精细胞数量减少等病理学改变以及细胞核固缩、染色质块状浓集、线粒体减少等超微结构改变均明显改善;睾丸组织凋亡细胞数量明显减少,细胞凋亡指数(AI)显著降低(P<0.05或P<0.01)。[结论]藏红花素能够有效抑制糖尿病大鼠睾丸组织损伤,其机制可能与其降低血糖作用有关。 |

Study on the protective effect of crocin on testicular tissue injury caused by diabetes mellitus |

[Objective] To investigate the protective effects of crocin on testicular tissue injury caused by diabetes mellitus.[Methods] The 100 SD rats were randomized according to body weight divided into normal control group,model group,Crocin[25,50,100 mg/(kg·d)] groups,the drugs were given by gastric administration for 12 weeks. The diabetic rat models were made by intraperitoneal injecting STZ[30 mg/(kg·d)] for 3d continuously. The general condition of the rats in every group was observed;the FBG,FINS,GHb were detected. The body weight was detected and the testis tissue index was calculated,the content of testosterone in serum was determined;the histopathological changes of testis tissue was observed by HE staining,the ultrastructure of testis tissue was observed by electron microscope,the apoptosis of testis tissue apoptosis was observed by TUNEL.[Results] After treatment with crocin 50~100 mg/(kg·d) for 12 weeks,the FBG decreased significantly,FINS increased significantly,GHb decreased significantly in type 2 diabetic rat models (P<0.05 or P<0.01);the body weight,the testis tissue index and the content of testosterone were significantly increased (P<0.05 or P<0.01). 12 weeks later,the ultrastructural changes such as atrophy of seminiferous tubules,loss of spermatogenic cells and reduction of spermatogenic cells in testis tissue of diabetic rats,and ultrastructural changes such as nuclear condensation,chromatin mass accumulation,and reduction of mitochondria were significantly improved;the number of apoptotic cells in the testis decreased significantly,the Apoptosis Index (AI) decreased significantly (P<0.05 or P<0.01).[Conclusion] Crocin can effectively inhibit testicular tissue damage in diabetic rats,which mechanism may be related to its role in lowering blood sugar. |
