基于数据挖掘和网络药理学的《临证指南医案·淋带》用药研究

摘要:
[目的] 挖掘《临证指南医案·淋带》的组方用药规律,探索治淋带基本组方的主要活性成分,初步研究治淋带基本组方的治疗机制。[方法] 收集《临证指南医案·淋带》的病案,采用TCMISS软件进行组方数据分析,探讨用药规律,挖掘核心药物及新处方,运用中药系统药理学数据库和分析平台(TCMSP)、中药网络数据库及分析系统(TCM-Mesh)、中药成分靶点数据库(HIT)、中药信息数据库(TCM-ID)、人类表型术语集(HPO)、DrugBank、毒性与基因比较数据库(CTD)及京都基因和基因组百科全书(KEGG)数据库分析治淋带基本组方各药物靶标间相互作用关系及相关通路。运用STRING探讨其相关性。[结果] 得到组方21则,药物54种,挖掘出2个核心药物组合和治淋带基本组方,得到治淋带基本组方的靶标基因1 192个,淋带的靶标基因52个,药物-疾病交集靶标基因10个,相关通路6条。[结论] 叶天士治疗淋带病早期以交通心肾为主,后期以滋补肝肾为主。药物作用机制可能为免疫炎症、激素调节等,同时受吞噬小体信号通路、NOD样受体信号通路的复杂影响。

Study on the medication of Guide to Clinical Practice with Medical Record: Lindai based on data mining and network pharmacology

Abstract:
[Objective] To explore the prescription medication rule in Guide to Clinical Practice with Medical Record:Lindai,explore the main active components of the basic prescription of lindai,and preliminarily study the treatment mechanism of the basic prescription of lindai.[Methods] TCMISS was used to analyze medication rules of prescriptions,explore the core drugs and new prescription. TCMSP,TCM-Mesh,HIT,TCM-ID,HPO,DrugBank,CTD and KEGG were used to analyze interaction relationship and related pathways among the drug targets in the basic prescription of lindai. The correlation were analyzed by STRING.[Results] The 21 prescriptions and 54 drugs were obtained,and 2 core drug combinations and basic prescription of treating lindai were excavated. The 1 192 target genes of basic prescriptions of treating lindai,52 target genes of lindai,10 target genes of drug-disease intersection and 6 related pathways were obtained.[Conclusion] In the early stage of treating lindai,YE Tianshi focuses on coordinating heart and kidney,while in the later stage,it focuses on nourishing liver and kidney. The mechanism of drug action may be immune inflammation,hormone regulation. It was affected by complex multi-targets and multi-pathways related with phagosome and NOD-like receptor signaling pathway.
