[目的]探讨川芎嗪、阿魏酸对心脏微血管内皮细胞白细胞黏附的影响。[方法]培养人心脏微血管内皮细胞,建立缺氧复氧模型,分离人外周血白细胞,计数法检测白细胞内皮细胞黏附率,反转录聚合酶链式反应(RT-PCR)检测内皮细胞黏附分子表达。[结果]缺氧培养2h,恢复正常培养1h,心脏微血管内皮细胞白细胞黏附率明显增加,川芎嗪、阿魏酸可降低缺氧复氧心脏微血管内皮细胞与白细胞黏附。缺氧2h复氧0.5h,细胞间黏附分子-1(ICAM-1),P选择素mRNA的表达较正常组升高,缺氧2h复氧4h,ICAM-1,P选择素,E选择素mRNA的表达较正常组升高。阿魏酸和川芎嗪均不同程度降低ICAM-1、P选择素、E选择素mRNA的表达。[结论]川芎嗪、阿魏酸可通过降低内皮细胞黏附分子表达,降低白细胞内皮细胞黏附,从而可能成为预防及治疗缺血再灌注损伤的有效方法。 |

[Objective] To study the effects of Ligustrazine and ferulic acid on leukocyte-cardiac microvascular endothelial cell adhesion.[Methods] Human cardiac microvascular endothelial cell (HCMEC) was cultured and hypoxia-reoxgenation model of endothelial cells were performd.The effect of Ligustrazine and ferulic acid on adhesion of leukocyte to activated cardiac microvascular endothelial cells was evaluated by counting.The effects of Ligustrazine and ferulic acid on E-selectin,P-selectin and ICAM-1 mRNA mRNA expression were determined by reverse transcription polymerase chain reaction.[Results] The adhesion rate of leucocyte to HCMECs was increased after hypoxia 2 h-reoxgenation 1 h.When endothelial cells were treated with Ligustrazine or ferulic acid ahead,the adhesion rate was decreased.The expression of P-selectin and ICAM-1 mRNAwere up regulated after hypoxia 2 h-reoxgenation 0.5 h.The expression of P-selectin and ICAM-1 mRNA were up regulated after hypoxia 2 h-reoxgenation 4 h.When endothelial cells were treated with ferulic acid (0.05 mmol/L,0.1 mmol/L) ahead,the expression of E-selectin,P-selectin and ICAM-1 mRNA were down regulated after hypoxia 2 h-reoxgenation 0.5 h.Ferulic acid 0.1 mmol/L can decrease the expression of P-selectin mRNA after hypoxia 2 h-reoxgenation 4 h.While ferulic acid 0.05 mmol/L can decrease the expression of E-selectin mRNA after hypoxia 2 h-reoxgenation 4 h.When endothelial cells were treated with Ligustrazine (0.0625mmol/L) ahead,the expression of E-selectin and P-selectin mRNA were down regulated after hypoxia 2 h-reoxgenation 0.5 h and the expression of E-selectin and ICAM-1 mRNA were down regulated after hypoxia 2 h-reoxgenation 4 h.[Conclusions] Ligustrazine and ferulic acid can inhibit the expression of E-selectin,P-selectin and ICAM-1 mRNA mRNA and leukocyte-endothelial cell adhesion.This maycontfibute toits protective effect against ischemia-reperfusion injury. |
