| [目的]观察疏肝导浊方药含药血清对肝癌细胞株(HepG2)细胞株的过氧化物酶体增殖物激活受体(PPARα),肝脂酶(HL)及载脂蛋白ApoB100,ApoAⅡ,ApoCⅡmRNA表达的影响,探讨其治疗脂肪肝的机制。[方法]以HepG2细胞株为研究对象,采用血清药理学方法,将含药血清分为高、中、低(16,8,4g/kg)剂量组、东宝肝泰对照组、模型组和空白对照组,研究疏肝导浊方药对HepG2细胞株PPARα及载脂蛋白的mRNA表达的影响。[结果]表明疏肝导浊方药含药血清能增加HepG2细胞PPARα、HL、ApoB100,ApoAⅡ,ApoCⅡmRNA的表达。[结论]疏肝导浊方药含药血清提高了PPARα、HL及载脂蛋白ApoB100,ApoAⅡ,ApoCⅡmRNA的表达,从而促进了肝细胞的脂代谢功能这可能是疏肝导浊方药治疗脂肪肝的重要机制之一。 |

| [Objective] To observe the effects of Shuogandaozhuo prescription on the expression of PPARα,HL and apolipoprotein B100,ApoAⅡ,ApoCⅡmRNA in HepG2 in vitro and explore the mechanism for treating fatty liver.[Methods] HepG2 Cells in vitro were chosen as the study object and the experiments were divided into six groups:the shugandaozhuo-containing serum groups high dose 16 g/(kg·d),middle dose 8 g/(kg·d),low dose 4 g/(kg·d),the Dongbaogantai group,the model group and the normal group.The experimental method was using the serology pharmacology.[Results] The shugandaozhjuo-containing serum could increase the expression of PPARα,HL and apolipoprotein B100, APOAII,ApoCⅡmRNA of hepatic cell line of HepG2.[Conclusion] It may be one of the mechanism for treating hyperlipemia syndrome and coronary heart disease. |
