银杏叶提取物EGB761对高胆固醇血症家兔血脑屏障通透性的影响*

摘要:
[目的]观察银杏叶提取物EGB761对高胆固醇血症家兔血脑屏障是否具有保护作用。[方法]以2%高胆固醇饲料喂养雄性新西兰大白兔,建立高胆固醇血症家兔模型。模型建立后,用50mg/(kg·d)EGB761灌胃3周作为EGB761组,以5mg(/kg·d)辛伐他汀灌胃3周作为阳性药辛伐他汀组。干预结束后,耳缘静脉给予25g/L伊文思蓝(EB),3h后麻醉,生理盐水心脏灌流后取材,通过比色法测定皮质、海马和嗅球EB含量。[结果]银杏叶提取物EGB761可以降低海马和嗅球EB含量,与模型组相比分别为P<0.01、P<0.05;与阳性药辛伐他汀疗效相比差异无显著性,P>0.05。[结论]银杏叶提取物EGB761能够降低由高胆固醇血症导致的血脑屏障通透性。

Effect of ginkgo biloba extract EGB761 on permeability of Blood-brain barrier in rabbits with hypercholesterolemia

Abstract:
[Objective] To observe whether Ginkgo biloba extract EGB761 can offer protection for Blood-brain barrier.[Methods] The male adult New Zealand rabbits were fed by 2% high cholesterol feed in order to establish the model of hypercholesterolemia in rabbitss. After that EGB761 was used to treat the rabbits at a dose of 50 mg/(kg·d) for 3 weeks and simvastatin, 5 mg/(kg·d), as a positive control drug . All the animals were injected by EB, 25 g/L, through auricle vein. 3 hours later, all animals were anaesthetized, and drawn sample after perfusion by normal saline. Finally, the content of EB in cortex, hippocampus and olfactory bulb was tested by means of colorimetric method.[Results] The content of EB in hippocampus and olfactory bulb was reduced by Ginkgo biloba extract EGB761, and they were P<0.01, P<0.05 compared with model group, which was not significant different compared to the effect of positive drug simvastatin (P<0.05).[Conclusion] Ginkgo biloba extract EGB761 can reduce Blood-brain barrier impairment caused by hypercholesterolemia.
