化痰通络法联合尿激酶溶栓对急性脑梗死大鼠不同脑区PAF、MCP-1的影响*

摘要:
[目的]通过观察化痰通络法联合尿激酶溶栓对急性脑梗死大鼠不同脑区血小板活化因子(PAF)和核细胞趋化蛋白-1(MCP-1)的影响。[方法]SD大鼠136只,随机分为5组,即正常组、假手术组、模型组、尿激酶组、化痰通络法联合尿激酶组,采用自身栓子法制备大鼠大脑中动脉栓塞模型,分别给予中药灌胃及尿激酶溶栓,Elisa法检测化痰通络法联合尿激酶溶栓对急性脑梗死后6h、72h、7d、14d脑组织PAF、MCP-1蛋白表达的影响。[结果]化痰通络法联合尿激酶溶栓能明显抑制PAF、MCP-1蛋白的表达。[结论]中药联合溶栓可减少溶栓后再梗死,其机制可能与抑制PAF、MCP-1表达有关。

Effect of huatan tongluo therapy combined with urokinase thrombolysis on PAF`MCP-1 in different brain regions in rats with acute cerebral infarction

Abstract:
[Objective] To observe the effect of Huatan(resolving phlegm) Tongluo(dredging collateral) therapy combined with urokinase thrombolysis on the PAF, MCP-1 in different brain regions in rats with acute cerebral infarction and investigate the action mechanism of recurrence of infarction of intravenous thrombolysis in rats.[Methods] 136 SD rats were divided into 5 groups randomly: normal group, sham operation group, model group, urokinase group, Huatan Tongluo Therapy combined with urokinase thrombolysis group. We made middle cerebral artery occlusion model in rats by using their own embolus, and treated by gavage with Chinese herbs and urokinase thrombolysis respectively. Then we observed the expression of PAF, MCP-1 in the rat brain tissue with ELISA method after acute cerebral infarction (6 h,72 h,7 d,14 d). [Result] Huatan Tongluo Therapy combined with urokinase thrombolysis could inhibit the expression of PAF, MCP-1 obviously.[Conclusion] Chinese herbs combined with thrombolysis therapy can reduce the probability of recurrence of infarction with intravenous thrombolysis, and the action mechanism might be related to inhibiting the expression of PAF and MCP-1.
