针刺对快速老化鼠线粒体钙离子浓度和活性氧簇的影响

摘要:
[目的] 探讨针刺对快速老化鼠线粒体钙离子浓度和活性氧簇的调节。[方法] 运用快速老化鼠SAMP8和正常老化鼠SAMR1作为实验对象,采用“三焦针法”(原“益气调血,扶本培元”针法),应用荧光指示剂及荧光探针标记,观察针刺对小鼠皮层、海马线粒体钙离子浓度以及活性氧簇含量的影响。[结果] SAMP8对照组皮层、海马线粒体钙离子浓度和活性氧簇与正常对照组相比,表达上调;针刺组表达下调并趋向正常组;非穴组表达上调,与针刺组有显着差异。[结论] 针刺能明显抑制线粒体内钙离子的蓄积,减少氧自由基的产生。非穴位的针刺在一定程度上会对机体产生不良刺激,加速氧化损伤。

Acupuncture influence to mitochondria calcium Ions’ concentrations and reactive oxygen species of SAMP8 mouse

Abstract:
[Objective] To discuss the acupuncture adjusting function to mitochondria calcium ions' concentration and reactive oxygen species of SAMP8 mouse. [Methods] Chose SAMP8 mouse and normal control SAMR1 as experimental subjects, with"tri-jiao acupuncture method" (the old name were nourishing-regulating qi acupuncture method) and marking method of fluorescent indicator and fluorescence probe, to observe the acupuncture influence to mitochondria calcium ions' concentrations of mouse' cortex hippocampus and reactive oxygen species. [Results] Combined with control group, mitochondria calcium ions' concentration of mouse' cortex and hippocampus in SAMP8 group was up-regulated, of which the acupuncture group was down-regulated to the control group and the non-acupoints group was up-regulated with significant difference. [Conclusion] It could obviously restrain the accumulation of mitochondria calcium ions and reduce the generation of oxygen radical by acupuncture. Meanwhile, acupuncturing at non-acupoints could produce harmful stimulation and accelerate oxidative damage.
