[目的] 测定经大孔树脂纯化后酸枣仁总皂苷的含量。[方法] 酸枣仁药材经乙醇提取,正丁醇萃取,大孔树脂纯化得酸枣仁总皂苷。以人参皂苷Rb1为标准品,香草醛-硫酸液显色,采用紫外分光光度法,543 nm为检测波长,以相应的吸收度A为横坐标(X),浓度(g/L)为纵坐标(Y)绘制标准曲线。[结果] 回归方程为:Ŷ= 32.526X - 1.675 9,r=0.999 7.皂苷测定在取量1.818~ 29.09 mg/L时,吸光度与皂苷含量呈良好线性关系。精密度实验相对标准偏差(RSD)为0.09%,重现性实验RSD为0.28%,样品和标准品稳定性实验RSD分别为0.42%和1.13%,加样回收率实验结果平均回收率为99.88%,RSD为1.13%,样品中总皂苷的平均含量为54.59%,RSD为0.38.[结论] 建立的含量测定方法可行,稳定,可以用于浸膏的质量控制。 |

Determination of total saponins from Semen Ziziphi Spinosae |

Abstract: |
[Objective] To determine the content of the total saponins from Semen Ziziphi Spinosae with purificated by macroporous resin.[Methods] The total saponins from semen Ziziphi spinosae were obtained by ethanol extraction, butanol extraction and macroporous resin purification. With ginsenoside Rb1 as standard, vanillin(sulfuric acid) as chromogenic agent, the ultraviolet spectrophotometry absorption wavelength was 543nm. [Results] The standard curve was linear in the range of 1.818 ~29.09 mg/L. The regression equation was Ŷ= 32.526X-1.675 9, r=0.999 7. The RSD of precision and recurrency were 0.09% and 0.28%, the RSD of control substance and testing substance in stability test were 0.42% and 1.13%, the average recoveries was 99.88%, RSD was 1.13%. The average content of total saponin in samples was 54.59%, RSD was 0.38.[Conclusion] The content determination method is feasible, stable, and can be used for quality control of extract. |
