[目的] 建立同时测定菟丝子中绿原酸、对-香豆素和金丝桃苷的含量的高效液相色谱法,为菟丝子的质量标准建立提供依据。[方法] 色谱柱为依利特ODS C18(4.6 mm×250 mm,5 μm);流动相为甲醇-0.05%甲酸水溶液,梯度洗脱,流速为1 mL/min,检测波长为360 nm。[结果] 在此色谱条件下,3种成分可完全分离。绿原酸、对-香豆素和金丝桃苷的线性分别为2.00~100 μg /mL(r=0.999 3),6.25~100 μg/mL(r=0.999 1),0.50~50 μg/mL(r=0.999 1)。平均回收率分别为100%(RSD=3.2%)、102%(RSD=1.7%)和102%(RSD=3.1%)。[结论] 该方法简便可行,重复性好,可用于菟丝子的质量评价。 |

[Objective] To establish a sensitive method for simultaneous determination of chlorogenic acid, p-coumarin and hyperin in order to provide some evidences for the quality control of Cuscuta chinensis Lam.. [Methods] Methanol and 0.05% formic acid were used as mobile phase. The sample was separated on an ODS C18 25 column(4.6 mm×250 mm,5 μm) by gradient elution. The flow rate was 1.0 mL/min. The wavelength was set at 360 nm. [Results] Chlorogenic acid, p-coumarin and hyperin were well separated by HPLC. Linearity ranges were 2.00~100 μg/mL(r=0.999 3), 6.25~100 μg/mL (r=0.999 1) and 0.50~5 0 μg/mL(r=0.999 1) for chlorogenic acid, p-coumarin and hyperin, respectively.[Conclusion] The simple and feasible HPLC method was established to evaluate the quality of Cuscuta chinensis Lam.. |
