阳和平喘颗粒对哮喘大鼠肺组织形态学及血清IL-17、TNF-α和IFN-γ表达的影响

摘要:
[目的]探讨阳和平喘颗粒(YHPC)对哮喘大鼠肺组织形态学及血清白介素-17(IL-17)、肿瘤坏死因子-α(TNF-α)和干扰素-γ(IFN-γ)表达的影响, 从抗炎和提高免疫功能方面探讨阳和平喘颗粒治疗支气管哮喘的作用机制。[方法]SD大鼠90只随机分为6组, 每组15只, 分别为正常组(N组)、模型组(M组)、阳和平喘颗粒低、中、高剂量治疗组(YHPC-L、YHPC-M和YHPC-H组)和地塞米松组(DXM组).其中, M、YHPC-L、YHPC-M、YHPC-H和DXM组均采用国际通用的方法, 即卵蛋白(OVA)腹腔注射致敏及联合雾化吸入激发建立哮喘模型, 同时, 应用氢化可的松进行腹腔注射以复制肾阳虚模型, N组使用生理盐水作为对照。苏木精-伊红(HE)染色检测各组大鼠肺组织病理学变化, 酶联免疫吸附法(ELISA法)检测各组大鼠血清IL-17、TNF-α和IFN-γ表达水平的变化。[结果]与N组相比, M组大鼠血清IL-17、TNF-α显着升高(P<0.01),IFN-γ明显降低(P<0.01);与M组相比, YHPC-L、YHPC-M、YHPC-H和DXM组大鼠血清IL-17、TNF-α均发生下降(P<0.01),IFN-γ则明显上升(P<0.01);与DXM组相比, YHPC-L、YHPC-M、YHPC-H组对IL-17和TNF-α的作用效果小于DXM(P<0.01),而对IFN-γ的作用效果强于DXM(P<0.01).[结论]阳和平喘颗粒可通过抗炎及增强机体免疫力而达到支气管哮喘标本兼治的目的。

Effects of Yanghe Pingchuan granules on the expression of IL-17, TNF-α and IFN-γ in rat asthma serum

Abstract:
[Objective] To investigate the effects of Yanghe Pingchuan granules on the expression level of IL-17, TNF-α and IFN-γ in rat asthma serum.[Methods] Ninty SD rats were randomly divided into six groups which were normal group (N group), model group (M group), low, middle and high-dose of Yanghe Pingchuan group (YHPC-L, YHPC-M and YHPC-H group) and dexamethasone group (DXM group). For inducing asthma, the rats were sensitized by intraperitoneal injection with ovalbumin(OVA), and then activated by inhalation. Furthermore, dexamethasone was applied to replicate kidney-deficiency rat asthma model, while the N group were treated with saline as a control. HE staining was used to evaluate the pathological changes of lung tissue in each groups, ELISA was used to detect changes of IL-17, TNF-α and IFN-γ in the rat serum. [Results] The expression of IL-17 and TNF-α is significantly higher in M group than in N group, but IFN-γ is obviously lower. Compared with M group, the levels of IL-17 and TNF-α decreased notably (P<0.01), while IFN-γ levels increased (P<0.01). The effect of Yanghe Pingchuan Granules on IL-17 and TNF-α is lower than DXM (P<0.01), whereas is stronger than DXM on IFN-γ(P<0.01).[Conclusion] Yanghe Pingchuan granules could cure bronchial asthma by anti-inflammatory, enhancing immunity and reducing pathological changes.
