甘草地上部分不同提取方法提取物成分HPLC-MS/MS分析

摘要:
[目的] 对甘草地上部分的水提物、醇提物和CO2超临界萃取物中黄酮类成分的组成进行分析。[方法] 高效液相色谱法(HPLC)Agilent SB-C18柱,乙腈-0.5%甲酸水梯度洗脱,体积流量1.0 mL/min,柱温30℃,进样量10 μL,检测波280 nm。质谱使用电喷雾离子源,负离子模式。[结果] 在水提物中共定性出9个化合物,主要为黄酮(醇)苷类物质;在醇提物中共定性出14个化合物,主要为黄酮(醇)苷元类物质和异戊烯基黄酮类物质;在CO2超临界萃取物中共定性出10个化合物,主要为异戊烯基黄酮类物质。[结论] 鉴定出甘草地上部分不同提取方法提取物中有不同性质的黄酮类物质,可以为后续提取富集研究提供依据。

Analysis on chemical constituents from extracts of different extraction methods of aerial parts of Glycyrrhiza uralensis by HPLC-MS/MS

Abstract:
[Objective] To analyze chemical constituents from water extract,ethanol extract and CO2 supercritical extract of aerial parts of Glycyrrhiza uralensis.[Methods] HPLC uses Agilent SB-C18 analytical column,acetonitrile-0.5% formic acid solution gradient elution,volume flow rate 1.0 mL/min,column temperature 30℃,injection volum 10 μL under 280 nm detection wavelength. Mass spectrometry uses electrospray ion source,negative ion mode.[Results] In water extract,9 compounds were identified mainly consists of flavonoids (alcohols) glycosides. A total of 14 compounds were identified in ethanol extract,mainly flavonoids (alcohols) aglycones and isopentenyl flavone. In CO2 supercritical extract,10 compounds were identified,mainly isopentenyl flavone.[Conclusion] Extracts of different extraction methods with different properties are acquired providing basis for further research.
