| [Objective] To establish the determination method of flavonoids compound in the extract of Folium Nelumbinis with HPLC.[Methods] The total quereetin content in the Folium Nelumbinis was determined with HPLC after hydrolysis.The procedure was as follows:the chromatographic column was Shim-pack CLC-ODS (4.6 mm×150 mm,5μm)g mobile phase was methanol-water-phosphoric acid (50:50:0.1)g flow rate was 0.8 mL/ming detection wavelength was 360 nm.[Result]The content of sophoretin showed a good linear relationship at the range of 0.013～0.13μg (R=0.999 9).In the extract of Folium Nelumbinis the average content of sophoretin was 4.94%,RSD=1.67%.The average recovery rate was 99.15%,RSD=0.60%.[Conclusion] This technique is a convenient and simple method with high accuracy.The method can be used for the determination of the total flavonoids in the extract of Folium Nelumbinis and for the controlling the quality of the total glycoside of it. |
