Determination of Aconitine alkaloids before and after compatibility of Radix Aconiti Lateralis preparata with HPLC

Abstract
    [Objective] HPLC was used to simultaneous determining the content of Mesaconitine,Aconitine and Hypaconitine in the water extract of Radix Aconiti Lateralis preparata,Radix Aconiti Lateralis preparata and Radix Ginger,Sini Decoction.The changes of the chemical composition in the water extract were studied to verify the compatibility theory of Chinese medicine.[Methods] Determination was performed by HPLC method,a waters Symmetry C18 (150 mm×4.6 mm.5 μm) was adopted with a mobile phase of methanol -0.1% triethylamine(60:40) solution at a flow rate of 1.0 mL/min;The detection wavelength was set at 235 nm and the column temperature was 35℃.[Results] The content of Hypaconitine was greatly increased after Radix Aconiti Lateralis preparata compatibility,suggesting that obvious increasing of X composition in the hydrolysate.Among them,the content of mesaconitine,aconitine was increased in Radix AeonitiLateralis preparata water extract with Radix Ginger.The content of mesaconitine was increased minimally and aconitine was greatly decreased in the sini Decoction.[Conclusion] The method was simple,precise and reliable for determination of aconitine alkaloids.It can illuminated the change rules of aconitine alkaloids before and after Radix Aconiti Lateralis preparata compatibility.
