Protective effects of Danhong injection on DPPH-induced injuries in the isolated hearts of rats |

Abstract
|
[Objective] To examine the protective effects of Danhong injection on DPPH-induced myocardial injuries in the isolated hearts of rats in Langendorff model. [Methods] The isolated hearts of rats were perfused in a Langendorff model for 30 min and pre-treatment with Danhong injection for 10 min. The hearts were subjected to Krebs-Henseleit(K-H)solution with DPPH 0.25 μmol/L for 10 min to cause myocardial dysfunction, and followed by 10 min recovery with K-H solution. The perfusion pressure(PP), left ventricular systolic pressure(LVSP), left ventricular end diastolic pressure (LVEDP), heart rate (HR), maximal and minimum rate of pressure development(±dp/dtmax)were recorded. And the heart rate pressure product (RPP)was calculated. [Results] The perfusion pressure significantly decreased after 10 min perfusion with Danhong injection. Perfusion with DPPH causes a remarkable decrease in RPP, +dp/dtmax and an increase in -dp/dtmax. Pretreatment with Danhong injection of 2.5 μL/mL and 5 μL/mL for 10 min significantly reduced the DPPH-induced myocardial injuries. [Conclusion] Danhong injection can dilate coronary artery and protect against the myocardial injuries induced by DPPH in isolated rat hearts. |
