The UPLC quality control method of Corydalis Rhizamo herbs and herbal pieces

Abstract
    [Objectiye] To develop a UPLC quality control method of Corydalis Rhizamo herbs and herbal pieces by determining the contents of active components.[Methods] The separation was performed on an ACQUITY UPLC BEH C18(100 mm×2.1 mm, 1.7 μm) column in the UPLC method. The mobile phases consisted of acetonitrile (B) and 0.1% formic acid aqueous solution(A, pH=5.0) with gradient elution. The flow rate and column temperature were set at 0.3 mL/min and 45 ℃, respectively. The injection volume was 5 μL. The contents of tetrahydropalmatine and dehydrocorydaline were taken as indexes for the water extract of corydalis rhizoma.[Results] The content of tetrahydropalmatine was in the range of 0.229 1~0.775 5 mg/g and that of dehydrocorydaline was 0.781 0~2.931 0 mg/g. The contents of tetrahydropalmatine and dehydrocorydaline varied greatly in different samples of Corydalis Rhizamo. [Conclusion] Tetrahydropalmatine and dehydrocorydaline were stable in the water extract, which could be used as the index components for the quality control of Corydalis Rhizamo. The UPLC method was fast, efficient and of higher sensitivity. It was suitable for the rapid detection of active components and the quality evaluation of traditional Chinese medicines.
