Abstract
[Objective] To investigate antihypertensive effect and possible mechanism of the Pineneedle Extract(PNE) in 2-kidney-1-clip(2k1c) hypertensive animal model. [Methods] Established the 2k1c hypertensive animal models. The rats were divided randomly into six groups: model group, western medicine group(Captopril), PNE high-dose group, PNE medium-dose group, PNE low-dose group and sham-operation group. Model group and sham-operation group both received 0.9% sodium chloride. Western medicine group received 1 kg/L Captopril suspensions. PNE high-dose group, PNE medium-dose group, PNE low-dose group received PNE solution, 5 kg/L, 3 kg/L, 2 kg/L, separately. Each group rats was given corresponding drugs by gavage for 4 weeks. Then monitor their blood pressure, measure serum nitric oxide(NO) and plasma endothelin(ET) content. [Results] Compared with model group, the PNE each groups rats of diastolic pressure(DBP) and systolic pressure(SBP) both reduced, especially SBP, which were statistically significant(P<0.05). The level of serum NO fell, while the level of plasma ET raised, which were statistically significant(P<0.05). Contrasted to western medicine group, the PNE each group rats of blood pressure decrease slowly. Both the decline of ET content and the rise of NO content were not statistically significant. [Conclusion] PNE can reduce blood pressure by adjusting endothelium-derived relaxing and contracting factor.
