| Abstract

| [Objective] To research the intervention effect of puerarin on alcoholic liver fibrosis in rats and the effects on the proliferation activation of hepatic stellate cell. [Methods] Eight-five male wistar rats were randomly divided into normal group, model, low, medium and high puerarin-dose and effective medicine group, a total of 6 groups. Except the normal group, alcoholic hepatic fibrosis model was produced in rats by giving gavage with alcohol supplemented by corn oil, pyrazol. After 12 weeks, the status of hepatic fibrosis was observed by HE and Masson stain, The expression of PCNA, α-SMA in liver tissue was detected by immunehistochemical method. The proliferation index of liver cell was calculated by the expression of PCNA. [Results] The degree of liver fibrosis of the model group and each medication administration group were significantly higher than the normal group, and the degree of liver fibrosis of each medication administration group was lighter than the model group. But the degree of liver fibrosis of the low-dose group was more serious than any other medication administration group .The proliferation index in the model group was higher than any other groups, and the proliferation index in normal group was lower than the other groups. The expression of α-SMA in the model group was higher than any other. The expression of α-SMA in the medium puerarin-dose group, the high puerarin-dose group and the effective medicine group were lower than the model group. There was no significant difference between the expression of α-SMA in the low puerarin-dose group and the model group. [Conclusion] The efficacy of peurarin in the prevention and treatment of alcoholic hepatic fibrosis are implemented by inhibiting the proliferation and activation of hepatic stellate cell in rats, and the effects of the medium and the high dose group are more significant. |
