Abstract
|
[Objective] To establish an UPLC method for simultaneous determination four index components such as gallic acid, ellagic acid, quercetin and kaempferol in Euphorbia Humifusa Willd. [Methods] A chromatographic separation method was established with Acquity UPLC BEH C18 column (100 mm×2.1 mm, 1.7μm) with methanol-acetonitrile (40:60, A)-0.2% formic acid (B) as mobile phases at the flow rate of 0.2 mL/min. The gradient elution was carried out as follows: 0~3 min, 5%A; 3~4 min, 5%~17%A;4~19min, 17%A;19~42 min, 17%~30%A; 42~50 min, 30%~34%A; 50~55 min, 34%~70%A. The detection wavelength was 270 nm at 0~4 min and 360 nm at 4~55 min, and the column temperature was 30℃. [Results] The four index components were well separated and each component had a good linear relationship in the liner range (r>0.999 7); and the recovery was in the range of 96.5%~102.9%. [Conclusion] The method is accurate, sensitive and repeatable for the comprehensive quality control of Euphorbia Humifusa Willd. |
