Effects of Qishen Yiqi pill on angiogenesis of cultured rat aortic ring

Abstract
    [Objective] To investigate the effect of Qishenyiqi pill on the angiogenesis of cultured rat aortic ring. [Methods] Three dimensional culture of rat aortic rings in 48 wells plate, divided them into four groups: normal group(culture with normal medium), VEGF group(culture with normal medium with 10 ng/ml VEGF), Qishenyiqi pill group(culture with normal medium with 25 μg/mL QiShenYiQi pills), and Qishenyiqi pill with VEGF group(culture with normal medium with 25 mg/L Qishenyiqi pills and 10 μg/L VEGF). Every group had three wells. Change the medium every three days. Record the number of the total number, total length, average length, the longest length of the microvessels which grow out of the aortic ring every two days from the 3rd day after culture until the neovascularization degradation. [Results] In the normal group, microvessels gradually grow out from the aortic ring in 1~3 days, and continue grow longer and longer in the 3~7 days, but the number of neovasculare was the same with day 3. Then number and length of the neovasculare decreased after 7 days, in the other word, the neovasculare gradually degraded. Compared with the normal group, the treatment group can promote angiogenesis germination (1~3 days) and growth in 3~7 days. But after 7 days, although the total number (except Qishen Yiqi pill group 13~15 days), total length and average length and the longest length of the new microvascular was increased than that of the normal group, but the new vascular rapidly degraded in the Qishen Yiqi pill group, in the VEGF group the number and length of the neovasculare was not obviously increased and the structure of new vessels in the day 11 was destroyed and degenerated and gradually, in the Qishen Yiqi combined the VEGF group the length of neovasculare was increased continully in 7~13 days, indicated that the angiogenesis in this group was increased in 7~13 days sustained. [Conclusion] Qishen Yiqi pills can promote angiogenesis of the three-dimensional culture of aortic ring angiogenesis, but this affection maintained for a short time, if combined with VEGF it can promote angiogenesis and maintain sustained growth for a long time, and delay the degradation of angiogenesis.
