| Abstract
|
| [Objective] To investigate the influence of Astragalus on expression of nuclear factor-κb(NF-κB) induced by high glucose in human proximal renal tubular epithelial cells(HK-2). [Methods] Human renal proximal tubular cells were cultured and divided into five groups: low glucose as control group; high glucose group; astragalus pretreatment group with concentration of 2 μg/mL, 20 μg/mL and 200 μg/mL. Each group has three samples. Culturing in 37 ℃ incubator for 24 hours, 48 hours and 72 hours, inverted microscope observe the cells. The expression level of NF-κB protein was detected by Western blotting. [Results] Compared with high glucose groups, the drug intervention groups grouped more better, expressed NF-κB protein level was lower(P<0.05), and the effect of astragalus were showed in dose-dependent manner. [Conclusion] High glucose may induce HK-2 cells to express NF-κB and decrease growth. Astragalus has protective effect to HK-2 cells by inhibiting HK-2 cells expression of NF-κB . |
