Abstract
|
[Objective] To study the mechanism of synergistic attenuating effect of Shenmai injection combined with oxaliplatin by observing the effect of Shenmai injection combined with oxaliplatin on transplanted tumors of colon cancer LOVO cells in nude mice.[Methods] An in vivo model of nude mice with LOVO cell transplantation was established. NC group, oxaliplatin group (8 mg/kg), combination group (oxaliplatin 8 mg/kg; Shenmai injection group (10 mg/g) and Shenmai injection group(Shenmai injection 10 mg/g) were set up. Administered by intraperitoneal injection. The tumor volume was recorded every other day from the date of administration. The growth curve was drawn. The effect of oxaliplatin on the tumor inhibition rate of the transplanted tumor was observed after 14 d of administration. Blood samples were taken after the mice were sacrificed, the spleen tissue was weighed to calculate the spleen index; the tumor tissue was removed. The expression of basic fibroblast growth factor (b-FGF) and platelet-endothelial cell adhesion molecule (CD31) in tumor tissue was detected by chemical method.[Results] The tumor inhibition rate of combination of Shenmai injection and oxaliplatin was 49%. Combination therapy could inhibit the growth of tumor tissues and had no obvious toxicity to the liver and kidney function of tumor-bearing mice. The combined treatment group could significantly reduce the expression of b-FGF and reduce the microvessel density (MVD) of tumor tissue.[Conclusion] Shenmai injection can reduce the effect of oxaliplatin on immune organs and liver function in mice. The mechanism of enhancing anti-colon cancer Oxaliplatin action may be related to the inhibition of angiogenesis signaling pathways mediated by b-FGF. |
