| [Objective] To evaluate the effect of Tongluo Zhitong formulation on histomorphology and innate immunity of synovium and cartilage in knee osteoarthritis of different degrees.[Methods] The KOA model of rats was established by Hulth method. The KOA model of rats was intervened by the method of ultrasound promoting Tongluozhitong formulation. The pathological changes of synovium and cartilage were observed to determine the degree of lesion, and the cartilage was evaluated by Mankin's score. The expression of TLR4, NF-κB and MyD88 in cartilage and synovium were observed by immunofluorescence double labeling technique.[Results] The injury degree of synovium and cartilage in the intervention group of Tongluo Zhitong formulation at 2, 4 and 8 weeks was significantly less than that in the control group. In innate immunity, the expression of TLR4, NF-κB and MyD88 in the intervention group of Tongluo Zhitong formulation was weaker than that in the control group.[Conclusion] Tongluo Zhitong formulation can inhibit knee osteoarthritis in different degrees, which proves that Tongluo Zhitong formulation can inhibit the occurrence and development of knee osteoarthritis by inhibiting the expression of TLRs/MyD88 signaling pathway related components. |
