Abstract
|
[Objective] To observe the regulation effect of Qingre Lishi Recipe on acid inositol 3-kinase (PI3K) pathway in fatty liver rats.[Methods] A total of 60 rats were randomly divided into 6 groups:normal control group,high fat model group,polyene phosphatidylcholine group,Qingre Lishi Recipe high dose group,Qingre Lishi Recipe medium dose group,and Qingre Lishi Recipe low dose group. The animal model of rat fatty liver was replicated by high-fat diet,and the heat-clearing and dampness group was intragastrically given 0.3 g/(100 g·d) (Qingre Lishi high-dose group),0.1 g/(100 g·d)(Qingre Lishi medium-dose group) and 0.05 g/(100 g·d)(Qingre Lishi low-dose group),and the polyene group was given 25 mg/(100 g·d). After 12 weeks,the material was taken. Pathological examined the degree of fatty liver in each group,by ELISA detected serum acute phase reaction protein (hs-CRP) content and ATPase activity,in biochemical analyzer detected fatty liver test group blood lipids (TC/TG),liver function (ALT/AST/GGT). Western-blot was used to detect phosphatidylinositol 3-kinase (PI3K)/Ark expression in fatty liver cells.[Results] The hs-CRP level in the high-fat model group was significantly higher than that in the high-dose group (P<0.05). The ATPase levels in each experimental group were decreased to different extents. The serum ATPase levels in the high-dose group were significantly different from those in the high-fat model group (P<0.05). The results of western-blot showed that the expression of PI3K/Ark in the hepatocytes of each experimental group existed significant differences,reduced expression of PI3K/Ark after treatment with Qingre Lishi recipe.[Conclusion] Qingre Lishi Recipe can improve the hepatocyte function recovery by improving the oxidative stress inflammatory mediators of ATPase,hs-CRP and PI3K/Ark hepatocytes,and can promote the recovery of liver cell function,at the same time,it also has certain effect of improving fatty degeneration. |
