Content determination of saikosaponins in Bupleuri Radix from different habitats |

Abstract
|
[Objective] To determine total saponins and four monomers saponins (saikosaponin a,d,c and f) in Bupleuri Radix from different habitats. [Methods] Using saikosaponin d as the standard,the total saponins in 24 batches of B. chinense DC. and 5 batches of B. scorzonerifolium Willd. were determined at 536 nm by UV-Vis spectrophotometry. The content of saikosaponin a,d,c and f were determined by HPLC-ELSD. [Results] The content of saikosaponin d in the same batch of samples was generally higher than that of saikosaponin a. There were significant differences in the contents of saikosaponin a,c,d and f from different habitats. There was a good linear relationship in range of 0.08~0.4 mg (r=0.999). The recovery of sample addition ranged from 99.2% to 103%,RSD=1.42%. The content of total saponins ranged from 5.32% to 13.9%,and the content of total saponins in B. scorzonerifolium Willd. was higher than that in B. chinense DC. The content of total saponins in B. scorzonerifolium Willd. from Linyi (Shandong Province) was the highest,reaching 13.9%. [Conclusion] The quality evaluation of Bupleuri Radix can not only determine the content of saikosaponin a,it should determine saikosaponins and saikosaponin a,c,d and f at the same time for more accurately. |
