Abstract
    [Objective] To investigated the pathology effect of PolygonumCuspidatum's effective components on kidneys of mice with diabetic nephropathy(DN).[Methods] KK-Ay mice with spontaneous type 2 diabetes were fed with high-fat diets for 3 weeks to establish DN models. The successful models of mice were randomly divided into the model group,the positive drug group,high-dose,medium-dose and low-dose groups of emodin and high-dose,medium-dose and low-dose groups of polydatin. The C57BL/6J mice were recruited as the control group. Each group included 6 mice. Each mice was administered once daily. After twelve weeks of intervention,all mice were sacrificed and the kidneys were harvested for hematoxylin-eosin (HE) and periodic acid-schiff (PAS) stain. The renal lesions were classified and scored under light microscope. The relative area of mesangium extracellular matrix was measured under the PAS stain. Ultrastructural morphological changes of renal tissue were measured under electronmicroscope.[Results] Compared with the model group,the renal tubular lesions in the high dose of polydatin group were significantly reduced (P<0.05);Mesangium extracellular matrix relative area was significantly decreased in polydatin group and the high dose group of emodin (P<0.01,P<0.05). The pathological changes of glomerulus were diminished in emodin group. The structure of podocytefootprocess was normal in the medium dose group of polydatin which can significantly reduce the ultrastructural lesions of renal tissue.[Conclusion] Emodin and polydatin can significantly lighten the renal lesions of mice with DN and have a protective effect on kidney.
