Effect of Qushi Tongluo Formulation on TNF-α and IL-6 expression in RAW264.7 infected with mycoplasma pneumoniae

Abstract
    [Objective] To observe the effect of Qushi Tongluo Formulation on TNF-α and IL-6 expression in RAW264.7 cells infected with mycoplasma pneumoniae (MP).[Methods] 1)Preparation of drug serum containing Qushi Tongluo Formulation:SD rats were randomly divided into blank control group,low,medium and high dose group of traditional Chinese medicine,and azithromycin group. Each group received continuous administration for 7days. 2) Establishment of MP-infected RAW264.7 cell inflammation model:model group and each treatment group were added with 100 μL of MP bacterial fluid,and blank control group was added with 100 μL of DMEM complete culture fluid,and cultured in incubator for 2 h. 3) The effect of Qushi Tongluo Formulation on the expression of TNF-aand IL-6 in MP infected RAW264.7 cells was detected by ELISA.[Results] Compared with the blank control group,the secretion of TNF-α and IL-6 in MP-infected RAW264.7 cells increased significantly (P<0.05). Compared with the model group,TNF-α and IL-6 were down-regulated in all groups (P<0.05).[Conclusion] Qushi Tongluo Formulation can downregulate the high expression of TNF-α and IL-6 in MP infected RAW264.7 cells in a dose-dependent manner,which may be one of the mechanisms by which Qushi Tongluo Formulation can reduce inflammation.
