Regulatory effect of curcumin on expression to relevant cytokines in colonic tissue about colitis mice

Abstract
    [Objective] To investigate the curative effect and possible mechanism of curcumin in different colon effect on colitis in mice.[Methods] The C57 mice randomly were divided into six groups,including the normal model group(saline),the model group(saline),curcumin low(50 mg/kg),middle(100 mg/kg) and high(200 mg/kg) dose groups,mesalazine group(300 mg/kg). After trinitrobenzene sulfonic acid (TNBS) was used to replicate the mouse colitis model,the mice were treated by 7 days to observe the general living habits of the rats such as weight,activity,hair gloss and fecal characters,the colonic mucosal injury score was observed and the pathological changes of the colon were observed by HE staining and the score was evaluated for curative effect. The level of GM-CSF,IL-2,IL-4,IL-12P40,IL-15,IL-21 and IL-23 were detected by ELISA.[Results] In the model group the levels of GM-CSF,IL-2,IL-12P40 and IL-21 in colonic supernatants increased,while the levels of IL-4,IL-15 and IL-23 decreased. Compared with untreated TNBS-induced colitis mice,the curcumin could reduce the expression of IL-2,IL-12P40 and IL-21 in the supernatant of colonic tissue of rats,and increase the levels of IL-4,IL-15 and IL-23.[Conclusion] The curcumin has therapeutic effect on colitis,and its possible mechanism is to regulate the cytokine balance and reduce inflammatory injury.
