Establishment of animal model of senile pruritus with blood deficiency and wind dryness syndrome and intervention effect of Zhufutang Ointment

Abstract
    [Objective] Based on the theory of "combination of disease with syndrome", we established and evaluated the senile pruritus(SP) animal model of blood deficiency and wind-dryness.[Methods] In this study, taking body weight as a block factor, 72 C57BL/6 mice were randomly divided into 6 treatment groups, namely blank group, model group, desonide cream group, Zhufutang Ointment low concentration group, Zhufutang Ointment medium concentration group and Zhufutang Ointment high concentration group, with 12 mice in each group. Adopting the idea of making model of disease before syndrome, in addition to the normal group, the other 5 groups used D-galactose Injection combined with the mixture of acetone and ether stimulation method to establish SP animal model, and then the low iron feed feeding combined with physical stimulation can successfully establish the SP animal model of blood deficiency and wind dryness. After modeling, the success of the model was verified by the general condition, apparent indicators, pathological index and biochemical index of mice.[Results] 1)General situation of mice:before modeling, mice in each group had good skin condition, bright eyes and good mental condition. Compared with the blank group, mice in the other groups appeared estless, irritable and dry stool after modeling(P<0.05). 2)Evaluation of apparent indicators:there was no significant difference in scratching behavior and water intake between each group before modeling. Compared with the blank group, the scratching behavior and water intake of the other groups were increased after modeling(P<0.05). 3)Pathological evaluation:after intervention, compared with the model group, the epidermal thickness and mast cell count of mice in the desonide cream group, Zhufutang Ointment low concentration group, Zhufutang Ointment medium concentration group and Zhufutang Ointment high concentration group decreased(P<0.05). 4)Biochemical index evaluation:after intervention, compared with model group, the level of TNF-α in mice of the desonide cream group, Zhufutang Ointment low concentration group, Zhufutang Ointment medium concentration group and Zhufutang Ointment high concentration group decreased (P<0.05), IL-4 cytokine and HYP level increased (P<0.05).[Conclusion] In this study, the animal model of SP blood deficiency and wind dryness syndrome was successfully established by using the idea of disease and then syndrome, D-galactose Injection combined with acetone and ether mixture stimulation, low-iron feed feeding and physical stimulation. The model has good stability and relatively strong operability, and is worthy of further application.
