Abstract
|
[Objective] To establish the UPLC fingerprint of Pinellia ternata,combined with chemical pattern recognition to screen the differential markers. [Methods] An ACQUITY UPLC HSS T3 column (2.1 mm×100 mm,1.8 μm) was used,the mobile phase was 0.1% aqueous phosphoric acid-acetonitrile,gradient elution,the flow rate was 0.2 mL/min,the injection volume was 10 μL,and the column temperature was 32℃,the detection wavelength is 260 nm. It was analyzed by a combination of similarity evaluation and chemical pattern recognition. [Results] Fifteen common peaks were identified from 18 batches of Pinellia ternata UPLC fingerprints,of which 16 batches of samples had similarities greater than 0.877,and the remaining 2 batches were 0.606 and 0.778 respectively,by cluster analysis,Pinellia ternata from different origins were divided into two categories. The results of principal component analysis and cluster analysis were consistent. The orthogonal partial least squares-discriminant analysis was used to screen out 6 differential markers,and three components (cytidine,hypoxanthine,guanosine) were identified by the reference substance. [Conclusion] The established fingerprint method is stable,reliable and repeatable. Combined with chemical pattern recognition,it can provide reference for quality evaluation of Pinellia ternata. |
