Abstract
|
[Objective] To rapidly identify the chemical components of Wendan Decoction by UPLC-Q-Exactive Orbitrap-MS. [Method] Waters Acquity BEH C18(2.1×100 mm,1.7 m) was used for gradient elution with 0.1% formic acid aqueous solution(A)-Acetonitrile(B) as mobile phase in positive and negative ion modes. The flow rate was 0.3 mL/min,the column temperature was 30℃,and the injection volume was 2 L. Adopting heated electrospray ion source(HESI),scan mode is Full MS/dd-MS2 and scan range m/z 100~1500. [Results] The compound was identified by comparing with the accurate relative molecular mass,multi-stage mass spectrometry fragment information,combining with literature reports and mass database information. A total of 121 ingredients were identified from Wendan Decoction,including 70 flavonoids,18 coumarins,9 saponins,11 nitrogen-containing ingredients,3 phenylpropanoids and 10 other components. Among them,15 components were confirmed after comparing with the reference substance. [Conclusion] UPLC-Q-Exactive Orbitrap-MS technology was used to systematically clarify the basic ingredients of Wendan Decoction,which can lay a foundation for the better research of the pharmacodynamic material basis and the reasonable formulation of quality standards. |
