Abstract
|
A method was developed for simultaneous determination of arginyl-fructose(AF) and arginyl-fructosyl-glucose(AFG) content in ginseng processing products. The determination was performed with PrevailTM C18 column(4.6 mm×250 mm,5 μm),mobile phase A:chromatographic acetonitrile,mobile phase B:5.0 mmol/L heptafluorobutyric acid in 0.7% trifluoroacetic acid solution,and the chromatographic conditions were 0% A(0 min);0% A(5 min);15% A(8 min);35%A(25 min),the flow rate was 0.8 mL/min,drift tube temperature was 115℃,gas flow was 3.2 L/min. The detection limits of AF and AFG were 0.015 and 0.010 mg/mL,respectively. The correlation coefficients of linear relationship are respectively AF:0.9997 and AFG:0.9999,indicating a good linear relationship. The relative standard deviations of precision,repeatability,stability and recovery of AF and AFG were 0.43%&0.37%,0.43%&0.55%,0.43%&0.49%,0.45%&0.15%,respectively. The recoveries of AF and AFG were 99.5% and 100% respectively,indicating that the method was stable. The content of AF in red ginseng,Chinese red ginseng and sun-cured ginseng was 0.74%,0.91% and 1.14%,and the content of AFG was 6.04%,5.08% and 0.85%,respectively. The method was successfully applied to the determination of AF and AFG in red ginseng,Chinese red ginseng and sun-cured ginseng. The content of AF and AFG in red ginseng was significantly higher than that in sun-cured ginseng. The advantages of this method were reliable measurement results,shorter measurement time and less interference from impurities. However,due to the small number of amino acids studied in this study,such as the variety of derivatives,it is still necessary to further investigate the separation degree. |
